50 µg specific antibody, lyophilized. Affinity purified with the immunogen. Rabbit serum albumin was added for stabilization. For reconstitution add 50 µl H2O to get a 1mg/ml solution of antibody in PBS. Then aliquot and store at -20°C until use.

Immunogen Synthetic peptide (aa 29 - 37 of rat GABA-A receptor α2 precursor protein) coupled to key-hole limpet hemocyanin via an added C-terminal cysteine residue.
Reactivity Human, rat, mouse; other species not tested yet.
Specificity Specific for GABA-A receptor α2.
Remarks
  • WB: The crude antiserum is highly recommended. This protein aggregates after boiling, making it necessary to run SDS-PAGE with non-boiled samples.
  • ICC, IHC: The affinity purified antibody is highly recommended for these applications.
    For best results use the protocol of Schneider Gasser et al., 2006.
  • ICC: This antibody is suitable for the surface staining of living cells. After washing cells with bound antibodies, they can be fixed and visualized with secondary reagents.

Control peptide for GABA-A receptor α2 antibodies
Cat. No. 224-1P 100 µg lyophilized peptide. For reconstitution add 100 µl H2O to get a 1mg/ml solution in PBS. Then aliquot and store at -20°C until use.
Control peptides should also be stored at -20°C when still lyophilized!
Recommended dilution Optimal concentrations should be determined by the end-user.
Remarks This control peptide consists of the synthetic peptide (aa 29 - 37 of rat GABA-A receptor α2 precursor protein) that has been used for immunization. It has been tested in preadsorption experiments and blocks efficiently and specifically the corresponding signal in Western blots. The amount of peptide needed for efficient blocking depends on the titer and on the affinity of the antibody to the antigen.

Remarks
  • WB: This protein aggregates after boiling, making it necessary to run SDS-PAGE with non-boiled samples.
  • ICC: This antibody is PFA fixation sensitive, use only mild fixation (2% PFA). Best results are obtained by application on living cells. After washing cells with bound antibodies, they can be fixed and visualized with secondary reagents.
  • IHC: For best results use the protocol of Schneider Gasser et al., 2006.
